Monoterpenes are the main components of essential oils and are possible precursors for many commodity chemicals and high energy density fuels. Monoterpenes are synthesized from geranyl diphosphate, which is also the precursor for the biosynthesis of farnesyl diphosphate. FPP biosynthesis diverts the carbon flux from monoterpene production to C15 products and quinone biosynthesis. In this study, we tested a chromosomal mutation of E. coli's native FPP synthase to improve GPP availability for the …Read more

  • Light-activated RNA interference in human embryonic stem cells
    with X. Huang, G. B. Braun, A. Pallaoro, D. P. Morales, J. Zasadzinski, D. O. Clegg, and N. O. Reich
© 2015.We describe a near infrared light-activated gene silencing method in undifferentiated human embryonic stem cell using a plasmonic hollow gold nanoshell as the siRNA carrier. Our modular biotin-streptavidin coupling strategy enables positively charged TAT-peptide to coat oligonucleotides-saturated nanoparticles as a stable colloid formation. TAT-peptide coated nanoparticles with dense siRNA loading show efficient penetration into a wide variety of hESC cell lines. The siRNA is freed from t…Read more

ROCK inhibition extends passage of pluripotent stem cell-derived retinal pigmented epithelium
    with R. H. Croze, M. J. de BuchholzRadeke, W. J. Thi, P. J. Coffey, and D. O. Clegg
Human embryonic stem cells offer a potentially unlimited supply of cells for emerging cell-based therapies. Unfortunately, the process of deriving distinct cell types can be time consuming and expensive. In the developed world, age-related macular degeneration is the leading cause of blindness in the elderly, with more than 7.2 million people afflicted in the U.S. alone. Both hESC-derived retinal pigmented epithelium and induced pluripotent stem cell-derived RPE are being developed for AMD thera…Read more

© AlphaMed Press.Somatic cells can be reprogrammed to an altered lineage by overexpressing specific transcription factors. To avoid introducing exogenous genetic material into the genome of host cells, cell-penetrating peptides can be used to deliver transcription factors into cells for reprogramming. Position-dependent C-end rule cell- and tissue-penetrating peptides provide an alternative to the conventional cell-penetrating peptides, such as polyarginine. In this study, we used a prototypic, …Read more
