The phosphatase CD148 promotes airway hyperresponsiveness through SRC family kinases
    with T. R. Katsumoto, M. Kudo, C. Chen, A. Sundaram, E. C. Callahan, J. W. Zhu, J. Lin, C. E. Rosen, B. N. Manz, J. W. Lee, M. A. Matthay, X. Huang, and D. Sheppard
Increased airway smooth muscle contractility and the development of airway hyperresponsiveness are cardinal features of asthma, but the signaling pathways that promote these changes are poorly understood. Tyrosine phosphorylation is tightly regulated by the opposing actions of protein tyrosine kinases and phosphatases, but little is known about whether tyrosine phosphatases influence AHR. Here, we demonstrate that genetic inactivation of receptor-like protein tyrosine phosphatase J, which encode…Read more

Distinct structural and catalytic roles for Zap70 in formation of the immunological synapse in CTL
    with M. R. Jenkins, J. C. Stinchcombe, B. B. Au-Yeung, Y. Asano, A. T. Ritter, and G. M. Griffiths
T cell receptor activation leads to a dramatic reorganisation of both membranes and receptors as the immunological synapse forms. Using a genetic model to rapidly inhibit Zap70 catalytic activity we examined synapse formation between cytotoxic T lymphocytes and their targets. In the absence of Zap70 catalytic activity Vav-1 activation occurs and synapse formation is arrested at a stage with actin and integrin rich interdigitations forming the interface between the two cells. The membranes at the…Read more

Combined Immunodeficiency Due to MALT1 Mutations, Treated by Hematopoietic Cell Transplantation
    with D. Punwani, H. Wang, A. Y. Chan, M. J. Cowan, J. Mallott, U. Sunderam, M. Mollenauer, R. Srinivasan, S. E. Brenner, A. Mulder, F. H. J. Claas, and J. M. Puck
© 2015, The Author.Purpose: A male infant developed generalized rash, intestinal inflammation and severe infections including persistent cytomegalovirus. Family history was negative, T cell receptor excision circles were normal, and engraftment of maternal cells was absent. No defects were found in multiple genes associated with severe combined immunodeficiency. A 9/10 HLA matched unrelated hematopoietic cell transplant led to mixed chimerism with clinical resolution. We sought an underlying cau…Read more

Modification by covalent reaction or oxidation of cysteine residues in the tandem-SH2 domains of ZAP-70 and Syk can block phosphopeptide binding (review)
    with P. R. Visperas, J. A. Winger, T. M. Horton, N. H. Shah, D. J. Aum, A. Tao, T. Barros, Q. Yan, C. G. Wilson, M. R. Arkin, and J. Kuriyan
© The Authors Journal compilation © 2015 Biochemical Society.Zeta-chain associated protein of 70 kDa and spleen tyrosine kinase are non-receptor tyrosine kinases that are essential for T-cell and B-cell antigen receptor signalling respectively. They are recruited, via their tandem-SH2 domains, to doubly phosphorylated immunoreceptor tyrosine-based activation motifs on invariant chains of immune antigen receptors. Because of their critical roles in immune signalling, ZAP-70 and Syk are targets fo…Read more
